NEPAGENE NEPA21 Type II Electroporator with CU500 Cuvette Chamber — MINT — TESTED — SEE VIDEO!

High-efficiency electroporation with programmable reverse-polarity pulses—without mandatory proprietary electroporation-buffer kits.

Offered is a Nepa Gene NEPA21 Type II Super Electroporator with CU500 Cuvette Chamber, in mint cosmetic condition and excellent working order. The NEPA21 combines separately adjustable poring pulses, transfer pulses, pulse-to-pulse voltage decay, and polarity reversal to help researchers optimize molecular delivery while preserving cell viability. It is designed for demanding applications involving mammalian cells, primary cells, stem cells, and other difficult-to-transfect biological samples.

Please watch the accompanying demonstration video, which shows the actual instrument powering on, accepting programmed settings, measuring impedance, executing pulse sequences, and displaying measured electrical output.

KEY ADVANTAGE: NO MANDATORY PROPRIETARY BUFFER KITS

One of the NEPA21’s most valuable features is its flexibility in buffer selection. Appropriate protocols can use commonly available culture media and saline solutions, including PBS, rather than requiring instrument-specific proprietary electroporation buffers. Bulldog Bio documents square-wave performance under low-resistance conditions representative of saline buffers. For mammalian-cell cuvette work, its commonly recommended medium is Opti-MEM.

For laboratories performing repeated transfections, optimization experiments, or larger numbers of samples, avoiding mandatory specialized buffer kits can substantially reduce recurring consumable expenses. The benefit continues beyond the initial purchase: users can develop protocols without committing every experiment to a particular instrument-specific buffer kit.

A common misperception about the electroporation of cells in suspension is that it requires costly specialized chemicals, running as much as $8 or more per sample. These additives can have unwanted effects on the cell and create another parameter which must be considered. Though electroporation has been around for many years, no protocols exist for high efficiency transfections without the use of such reagents. That is, not until the NEPA21. NepaGene has created THE most sophisticated apparatus for delivering DNA, RNA, and proteins into cells. Your cells stay in standard media, so all that’s required are the reusable electrodes. With a combination of low voltage short-and-long pulses and unique polarity reversal, the NEPA21 can transfect a huge variety of cells, tissues, and even organisms with amazing efficiencies. This patented pulsing technology has been shown to be both superior to and far more flexible than other techniques. And because most living cells are not bothered by low voltages, and transfections can be done in standard growth media, cytotoxic effects are nearly eliminated.

The included CU500 chamber uses conventional electroporation cuvettes; Bulldog Bio recommends standard 2 mm-gap cuvettes for its usual CU500 workflows. Disposable cuvettes and experimental reagents are purchased separately.

HIGH-EFFICIENCY DELIVERY WITH REVERSE-POLARITY PULSES

Rather than limiting the user to a single pulse type, the NEPA21 separates membrane permeabilization from the subsequent molecular-transfer process.

Pulse function	Purpose
Poring pulses	Relatively higher-voltage, shorter pulses designed to create transient membrane pores.
Reverse-polarity poring pulses	Optional polarity exchange adds flexibility when optimizing the permeabilization sequence.
Transfer pulses	Lower-voltage, longer pulses promote movement of charged molecules into permeabilized cells.
Reverse-polarity transfer pulses	Reverses the transfer direction and can improve transfection efficiency.

Programmable voltage decay provides another way to tailor successive pulses. Together, these controls allow the user to balance transfection efficiency and cell survival, rather than simply increasing electrical exposure. Nepa Gene describes this as its four-step multiple-pulse electroporation approach.

DETAILED PULSE CONTROL AND ELECTRICAL MONITORING

The NEPA21 gives the researcher direct access to the principal pulse parameters: voltage, pulse length, pulse interval, pulse count, decay rate, and polarity, with separate settings for poring and transfer pulses.

Its built-in monitoring provides more information than the programmed settings alone:

Pre-pulse impedance measurement helps assess the electrical load before treatment.
Measured voltage, current, and energy readouts document the delivered pulse sequence.
Separate setting and measurement displays make it convenient to review programmed conditions and instrument-reported output.

These features support method development, troubleshooting, and reproducibility. Bulldog Bio’s programming guide specifically recommends recording impedance and electrical output when optimizing experiments.

CONDITION AND FUNCTIONALITY

The instrument is in mint cosmetic condition and excellent working order. The protective film remains on the control panel and can be removed.

It has been function-tested in our laboratory. The demonstration video shows:

Power-on and the startup self-test sequence.
Selection and adjustment of poring and transfer parameters.
Programming of polarity reversal and voltage decay.
Sample impedance measurement.
Execution of pulse sequences and review of the instrument’s measured voltage, current, and energy values.

Electrical Testing and Biological Demonstration

In addition to the electrical functional testing shown in the video, this NEPA21 Type II was used in our laboratory for electroporation tests with mouse embryonic fibroblasts (MEFs) and mouse oocytes in the presence of fluorescent dextran-TRITC. Representative microscopy images from these tests are included in the listing.

The photographs show the actual NEPA21 and CU500 chamber being offered, including the chamber contacts, cables, connectors, and instrument identification label.
